Application
Der Phosphatasehemmer-Cocktail verbessert die Ausbeute an intakten Proteinen, indem er Inhibitoren zu Enzymen hinzufügt, die Proteine in Zellextrakten modifizieren. Dieses Produkt wurde an Extrakten von Tiergeweben wie humanem und bovinem Lebergewebe sowie an Extrakten von A431-, CHO- und U937-Zellen getestet. Es wurde optimiert und auf L-Isoenzyme von alkalischer Phosphatase mit pNPP sowie auf Serin-Threonin-Proteinphosphatasen getestet.
Biochem/physiol Actions
Dieses Gemisch enthält Einzelkomponenten mit spezifischen Hemmeigenschaften. Cantharidin hemmt die Proteinphosphatase 2A. (-)-p-Bromolevamisol hemmt L-Isoformen alkalischer Phosphatasen. Calyculin A hemmt die Proteinphosphatasen 1 und 2A.
Preparation Note
Dieser Cocktail wird als klare Lösung in Dimethylsulfoxid geliefert. Es wird empfohlen, den Cocktail auf 1 ml Cocktail pro 500 mg Proteinextrakt aus Gewebe oder Zellen zu verdünnen. Oftmals wird eine Endkonzentration von 1 % (v/v) verwendet, die Phosphataseaktivitäten im 27.000 x g zentrifugierten Überstand aus humanem Plazenta- und bovinem Lebergewebe sowie im 100.000 x g zentrifugierten Überstand von A431-, CHO- und U937-Extrakten hemmen.

This phosphatase inhibitor cocktail does not contain a chelator. A phosphatase inhibitor cocktail is not offered that includes EDTA at this time. Other protease inhibitor cocktails with chelators are offered.
