General description

Indole is the primary product of tryptophan breakdown by tryptophanase. Structurally, it is characterized by a benzene ring fused to a pyrrole ring, containing a five-membered nitrogen-containing ring. This simple structure forms the core of a vast array of natural and synthetic compounds with a wide range of biological activities. In bacteria, indole is a critical signaling molecule, produced as a metabolite during the breakdown of the amino acid tryptophan. It plays a pivotal role in bacterial behavior, influencing biofilm formation, spore germination, and the expression of virulence factors. In the context of metabolism in larger organisms, indole derivatives are integral to the functioning of the central nervous system, serving as precursors for the synthesis of neurotransmitters such as serotonin and melatonin. These compounds are consequential in regulating mood, appetite, sleep, and circadian rhythms. Indole metabolism in the gut microbiota has been implicated in maintaining the integrity of the intestinal epithelial barrier and modulating the immune response, reflecting the complexity and importance of this molecule in biological systems.

Biochem/physiol Actions

The indole assay kit is based on a modified version of Ehlrich′s and Kovac′s reagents, which reacts with indole to produce a colored compound at 565 nm. The intensity of this colored compound is directly proportional to the indole in the sample. Linear detection range from 3 to 100 μM indole in 96-well plate assay.

  1. Can this kit be used with human serum, plasma, or fecal samples?

    1. There is interference in serum samples due to precipitation. This issue can be greatly reduced by deproteinating with a filter, but some interference may still occur. This kit has not been validated for use with plasma samples or fecal samples. The reagent used in this kit reacts with urobilinogen, which is also excreted in feces. The kit would be unable to distinguish between indole and urobilinogen.

  2. If the test assumes it's indoxyl sulfate, a high conc. say 80-90 um would indicate toxicity, considering its a uremic toxin? Higher ILA/IPA on other hand would mean a beneficial indole.

    1. This kit is not intended to categorize a test sample as toxic. The test is designed to measure urinary Indican (Indoxyl sulfate) and assumes that most of the Indole in the sample exists as urinary Indican (Indoxyl sulfate). The test does not distinguish between Indole, Indican, or urinary Indican (Indoxyl sulfate), so it will likely detect other species such as Indolepyruvic acid, Indolelactic acid, and Indolepropionic acid if the reactive carbon is available. The test does not convert the indole derivative before the DMAB reaction.

  3. Is any up front extraction of the biological sample necessary before running the assay (example: solvent extraction)?

    1. The necessity for extraction often depends on the type of biological sample (e.g., serum, plasma, tissue, or cell lysates). Solid tissues may require extraction, while fluid samples may not.

  4. Is this working in 560nm in microplate reader?

    1. For optimal results, the plate should be read at 565 nm, as per the kit protocol.

  5. Will this kit react with tryptophan alone. For example will it react in the presence of 100µM Trp?

  6. Does this kit only measure indole, or does it detect a wide variety of indole-containing compounds such as indolepyruvic acid, indolelactic acid, and indolepropionic acid?

    1. The kit has not been tested with those compounds. The test is specifically designed to measure urinary indican (indoxyl sulfate), assuming that most of the indole in the sample exists as urinary indican (indoxyl sulfate).

  7. Does this assay detect both indole and all its derivatives? Or is there a specificity towards some indoles?

    1. Unfortunately, this kit has not been tested with Indole compounds such as indolepyruvic acid, indolelactic acid, and indolepropionic acid. The test is designed to measure urinary indican (indoxyl sulfate). It assumes that most of the indole in the sample exists as urinary indican (indoxyl sulfate). The test does not distinguish between indole, indican, or urinary indican (indoxyl sulfate) so it will likely pick up the species enumerated if the reactive carbon is available. The test does not convert the indole derivative before the DMAB reaction.
