General description

Die auch als Sialidase bezeichnete Neuraminidase ist ein Enzym, das terminale Sialinsäurereste auf Polysacchariden-Ketten hydrolisiert. Sie wird hauptsächlich in Mikroorganismen wie Bakterien und Viren exprimiert. Es wird angenommen, dass die Spaltung von Sialinsäureresten durch Neuraminidase bei der Infektion durch Gruppeviren mehrere Rollen spielt. Weiterhin wird davon ausgegangen, dass sie die Penetration von Schleimhäuten, den Befall von Zielzellen, die Elution von Nachkommenviren aus infizierten Zellen und die Prävention einer Selbstaggregation unterstützt. Daher ist die Neuraminidase ein wichtiges Ziel für die Entwicklung von Influenza-Wirkstoffen und für einfache, direkte und automatisierungsbereite Verfahren für das Messen der Neuraminidase-Aktivität in breitgefächerten Anwendungen im Forschungsbereich und bei der Wirkstoffentwicklung

In diesem Test wird die Neuraminidase-Aktivität mithilfe eines Enzym-Assays bestimmt, bei dem von der Neuraminidase freigesetzte Sialinsäure in einem kolometrischen (570 nm)/fluorometrischen (λex = 530/λem = 585 nm) Produkt resultiert, das zur Neuraminidase-Aktivität in der Probe direkt proportional ist. Der lineare Detektionsbereich bei 37 °C beträgt 0,1–10 Einheiten/L für kolorimetrische Assays und 0,01–2 Einheiten/L für fluorometrische Assays.

  1. Is the standard provided (MAK121C) sialic acid? What is the catalytic function of the enzyme (MAK121D)? If the sample is serum, is it recommended to heat-inactivate it before the assay? Thank you.

    1. The standard in MAK121C is sialic acid. MAK121D enzymes catalyze reactions that convert sialic acid to pyruvate, which then reacts with a dye reagent to yield a quantifiable color signal.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   5. Is it possible to use the Neuraminidase Activity Assay Kit (MAK121-1KT) for testing bacterial cultures, given that references indicate the use of plasma, serum, and purified enzymes as samples?

    1. The kit has been verified for use with bacterial cultures. Although there isn't a tailored sample preparation protocol for bacterial cultures, the following general cell lysate preparation method is suggested:
      To prepare cell samples, first, suspend approximately two million harvested cells (2 × 10^6) in 400 μL of PBS on ice. Cell lysis can be accomplished by either using a Dounce homogenizer for 10-20 passes while on ice or through sonication in an ice-water bath. The effectiveness of lysis should be verified under a microscope. After lysis, centrifuge the homogenate at 14,000 g for 10 minutes and then transfer the clear supernatant to a clean tube. It is recommended to run a pilot assay with different sample dilutions to identify a dilution that falls within the linear range of the standard curve for accurate assays. If immediate analysis is not possible, most samples can be preserved at -80°C.

    1. The substrate utilized is fetuin, which is a heavily sialylated protein derived from fetal bovine serum. The specific type of sialic acid linkage present in this substrate has not been characterized. However, fetuin typically exhibits two types of linkages: α2,3- and α2,6-linkages. It is more common for α2,3-linkages to be present in fetuin substrates, which suggests that experiments targeting this linkage type are likely to be successful. Nonetheless, it is not possible to provide a definitive statement on the exact type of linkage present.
