General description

Glycerin, auch „Glyzerin“ oder „Glycerol“ genannt, ist ein dreiwertiger Zuckeralkohol und integraler Bestandteil von Fettsäuren. Darüber hinaus ist es in Phospholipiden und Triglyceriden zu finden. Glycerin ist eine süß schmeckende, hygroskopische Flüssigkeit. Glycerin wird häufig in Nahrungsmitteln, Getränken und pharmazeutischen Formulierungen verwendet und ist das Hauptabfallprodukt der Herstellung von Biodiesel durch Umesterung.

Application

Das Glycerin-Assay-Kit wird verwendet, um die Glycerinfreisetzung in Überständen von 3T3-L1-Zellkulturen der Maus zu quantifizieren und um den Glycerinspiegel im Plasma zu messen.
Dieses Kit ist geeignet für den Glycerinnachweis in Nahrungsmitteln, Getränken, pharmazeutischen Formulierungen und biologischen Proben.

Biochem/physiol Actions

In diesem Kit wird die Glycerinkonzentration durch einen gekoppelten Enzym-Assay bestimmt, der Glycerinkinase und Glycerin-Phosphatoxidase enthält und proportional zum vorhandenen Glycerin ein kolorimetrisches (570 nm) bzw. fluorometrisches (lex = 535/lem = 587 nm) Produkt ergibt. Die lineare Detektionsbandbreite dieses Kits liegt bei 10–1.000 μM (kolorimetrisch) und 2–50 μM (fluorometrisch).

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. Is this assay (MAK117) and the free fatty acid assay kit (MAK466) suitable for use with serum or supernatant samples which have been frozen prior to use?

    1. Yes, the serum or supernatant samples can be stored frozen. The glycerol analyzed by the MAK117 kit and the fatty acids analyzed by the MAK466 kit would not be harmed or their concentration changed by freezing samples. In fact, if samples need to be stored, they would logically be more stable if kept frozen.

  3. Does the presence of ethylene glycole, methanol or propanediols affect the analysis?

    1. There is no information regarding possible interference of Ethylene glycol, Methanol, or Propanediols with the assay kit, but such interferences are not likely. The use of Glycerol kinase makes the kit specific to the reaction of Glycerol with ATP.

  4. I'd like know what is the maximum concentration of glycerol measured by the instrument? How precise is the analysis measurement (standard deviation expected)? Can it be used for a water solution at 5% glycerol in weight (glycerol 99% pure)?

    1. Linear detection range in 96-well plate: 10 to 1000 µM (92 µg/dL to 9.2 mg/dL) glycerol for colorimetric assays and 2 to 50 µM for fluorimetric (1.84 µg/dL - 46 µg/dL) assays. A 5% glycerol solution with a density of approximately 1.01 g/mL translates to a solution with a concentration of 0.548 M or 548 mM or 548,000 µM. The sample would have to be diluted at least 1000x before it falls into the middle of the linear detection range for the colorimetric assay. Please dilute the sample in water and account for the dilution in the glycerol concentration calculation. No specific upper limit of quantitation or standard deviation has been assigned using this kit for this sample.

  5. After reading the description of this product, I can't help but wonder what the contents of the kit are, and if there is any additional documentation related (protocol example, enzyme description, standard concentrations and curves).

The formulation of the components is considered proprietary, with the exception of the standard - 100mM Glycerol. This is an enzymatic assay that utilizes glycerol kinase and glycerol phosphate oxidase. Please see the link below to review additional details:

  6. Is there any information available on how to prepare tissue for assay, considering that technical bulletin MAK117 does not include any recommendations on this matter?

    1. The general recommendation for tissue sample preparation is as follows:
      1. Begin with 20-100 mg tissue and add 200-1000 uL ice-cold PBS.
      2. Lysis can be achieved by homogenization (10-20 passes in a Dounce homogenizer on ice) or by sonication (preferably performed in an ice-water bath). The degree of tissue lysis can be checked under a microscope.
      3. Centrifuge the homogenate at 14,000 g for 10 min.
      4. Transfer the clear supernatant into a clean tube and use it for the assay.

  7. Can serum glycerol content be analyzed or just plasma?

    1. Yes, this product can determine glycerol content in serum samples.

  8. How long are reagents stable at 2-8 C after initial thaw?

    1. The Assay Buffer, MAK117A, may be stored at 2–8 Celsius. For all other components, it is recommended to aliquot and freeze at - 20 deg C. Avoid repleated freeze/thaw cycles.

  9. How is shipping temperature determined? And how is it related to the product storage temperature?
