General description

Die als Trockennährmedium erhältliche Luria-Bertani (LB)-Bouillon wird als Lennox LB bezeichnet und verfügt über Aminosäuren als Kohlenstoffquelle.
Lennox LB ist ein gerne genutztes Medium für das Mikrobenwachstum, das auch für die Kultivierung von E. coli verwendet wird. Die nährstoffreiche mikrobielle Bouillon enthält Peptide, Aminosäuren und Kohlenhydrate in einer Formulierung mit niedrigem Salzgehalt.

Application

Die LB-Bouillon (Lennox) wird für die Kultivierung von Escherichia coli und Bacterial Artificial Chromosomes (BACs) verwendet.
Geeignet für die nicht selektive Kultivierung von E. coli-Stämmen für das Klonen, die DNA-Plasmidproduktion und die Produktion rekombinanter Proteine. Ebenso geeignet für die selektive Kultivierung bei entsprechender Zugabe von Antibiotika, einschließlich derer, die Lösungen mit geringem Salzanteil erfordern, wie Zeocin®.

Preparation Note

Je eine 1,1-g-Tablette in 48,3 ml Wasser geben (kein Rühren erforderlich). Zur Sterilisierung 20 Minuten lang bei 121 °C autoklavieren. Bei Bedarf vor der Zugabe weiterer Stoffe, z. B. Antibiotika, abkühlen lassen.

  1. How long can these tablets be stored at room temperature in the bottle (after opening).

    1. This product is assigned retest date of approximately 3 years from the date of Quality Release. This date is lot-specific and may be found in the product Certificate of Analysis. When stored at room temperature, in a tightly sealed container, the material will be stable at least until the assigned retest date.

  2. Do the tablets contain any additives to adjust the pH of the media? Also, what should the final pH of the media be?

    1. The pH of the media can be 7.0, but depending on the application, it can range from 7.0 to 8.0. The pH can be adjusted using 1N NaOH and 1N HCl.

  4. Which bacterial culture medium is the best choice for my application?

    1. Each of the broths will likely grow E. coli very well, but there are still general guidelines for choosing a broth when you are working without a protocol. Generally:LB - Miller and LB - Lennox are used for E. coli growth and maintanence, DNA plasmid production and protein production.  The Lennox formulation has a lower salt content required for some salt-sensitive selection antibiotics.LB - Luria low salt is used for special applications where the E. coli growth or other constraints require the lowest possible salt content.Terrific Broth is used for higher yield protein production and high yield DNA plasmid production, because of the faster growth of the E. coli in this medium.SOB is used for protein production, DNA plasmid production and the generation of high-efficiency competent cells.SOC is used for initial growth of competent cells and the transformation procedure.

  5. Will adding magnesium to the culture medium increase cell density?

    1. In microbial broth formulations that do not already contain magnesium, the addition of 10-20 mM MgCl2 or MgSO4 may increase cell densities.  You may need to also increase the shaking speed of the incubator.

  6. What are the differences among the Luria, Lennox and Miller LB formulations?

    1. LB, (originally termed lysogeny broth) was initially composed of tryptone, yeast extract, NaCl and glucose.  Soon after, the glucose was omitted (Miller's version), and later the NaCl content lowered by half (Lennox's version).  For some applications, even lower salt is required (Luria's low salt version).
