Allgemeine Beschreibung

Cyclooxygenase 1 is a 71 kDa membrane bound protein predominantly present in endoplasmic reticulum. It has three domains, the epidermal growth factor (EGF) like domain, enzymatic and membrane binding domain.[1] Cyclooxygenase 1 mediates prostaglandin synthesis and is modulated by anti inflammatory nonsteroidal drugs.[2]

Anwendung

Cyclooxygenase 1 from sheep has been used as positive control protein in western blot analysis of osteoarthritis samples[3] and in prostaglandin synthase activity assay.[4]

Biochem./physiol. Wirkung

COX-1 catalyzes the conversion of arachidonic acid to prostaglandin H2 (the first step in the biosynthesis of prostaglandins, thromboxanes, and prostacyclins). It is involved in the homeostatic role of eicosanoids and constitutively expressed in almost all animal tissues. Has an apparent KM of 8.3 μM for arachidonic acid.

Einheitendefinition

One unit consumes one nanomole of oxygen per minute at 37 °C in 0.1 M Tris-HCl buffer, pH 8, containing 100 μM arachidonate, 5 mM EDTA, 2 mM phenol, and 1 μM hematin.

Physikalische Form

Solution in 80 mM Tris-HCl, pH 8, with 0.1% TWEEN® 20, 300 μM diethyldithiocarbamate and 10% glycerol.
