General description

Molekülmasse: 51–54 kDa.
α-Amylase, isoliert aus Schweine-Pankreas, ist ein Glykoprotein. Sie ist eine einzelne Polypeptidkette von ~475 Resten, die zwei SH-Gruppen und vier Disulfidbrücken sowie dicht gebundenes Ca2+ für die Stabilität enthält. Chloridionen sind notwendig für Aktivität und Stabilität. Der pH-Wertbereich für Aktivität beträgt 5,5 bis 8,0, wobei der optimale pH-Wert bei 7 liegt.

Application

α-Amylase wird zum Hydrolysieren von α-Bindungen von α-verbundenen Polysacchariden wie Stärke und Glykogen eingesetzt. Produkt A6255 stammt aus der Schweine-Pankreas und ist vom Typ I-A. α-Amylase von Sigma wurde in verschiedenen Pflanzenstudien, z. B. Metabolismusstudien in Arabidopsis, eingesetzt[1].

Biochem/physiol Actions

α-Amylase hydrolysiert die α-(1,4)-Glucanbindungen in Polysacchariden mit drei oder mehr durch α-(1,4) verbundene D-Glucose-Einheiten. Natürliche Substrate wie Stärke und Glykogen werden zu Glucose und Maltose gespalten. α -Amylase aus Schweine-Pankreas ist ein Glykoprotein, das aus einer einzelnen Polypeptidkette mit etwa 475 Resten besteht, die zwei SH-Gruppen und vier Disulfidbrücken sowie dicht gebundenes Ca2+ für die Stabilität enthält.

    1. The α-Amylase product, this product, contains 10 mg of protein, referring specifically to the protein amount rather than the total solid. The concentration of mg protein/mL is lot-dependent. Using lot 0000367139 as an example, the Certificate of Analysis indicates a protein concentration of 24 mg/mL in the saline suspension. For this 10 mg of protein, the volume is approximately 0.417 mL. If the suspension contains 1 mL, the activity, based on this lot’s 1162 units/mg protein, should be about 27,888 U/mL (24 mg/mL × 1162 units/mg = 27,888 U/mL, not 10,000 U/mL).

  2. A 6255 is declared with an activity of 1000 U/mg. 10 mg of enzyme contain 10000 units or not? What is the amount of liquid in the pack?

    1. This α-Amylase is sold based on mass, and is guaranteed to have a specific activity that is equal to or greater than 1000U/mg. Therefore, the 10mg size will have a minimum of 10,000 units.

  3. I have A6255 enzyme of 10 mg. If I dissolve it with 1 ml of buffer, it means that the activity is about 10000 U/ml. What I mean is that it contains 10 mg of solid or 10 mg of protein?

    1. The activity of this product, ?-Amylase from porcine pancreas, is batch specific. An example batch 0000367139 has 24 mg protein/ml (UV) and 1162 units/mg protein. Therefore, a 10 mg vial from this batch would have 11,620 units.
