General description

Spermidin ist ein Polyamin und ein trivalentes organisches Kation.Spermidin ist eine natürlich vorkommende Polyaminverbindung mit verschiedenen biochemischen Eigenschaften. Es spielt eine wichtige Rolle bei der Genexpression und ist an DNA-Replikation, -Transkription und -Translation beteiligt. Spermidin ist von entscheidender Bedeutung für Zellwachstum, -proliferation und -differenzierung und hat Auswirkungen auf die Progression des Zellzyklus und die Gewebeentwicklung. Es besitzt eine antioxidative Aktivität und schützt die Zellen vor oxidativem Stress, indem es freie Radikale abfängt. Spermidin stimuliert auch die Autophagie und fördert den Abbau von geschädigten Organellen und Proteinaggregaten für die zelluläre Homöostase. Mit neuroprotektiven Effekten unterstützt es das neuronale Überleben, die synaptische Plastizität und schützt vor neurodegenerativen Erkrankungen. Darüber hinaus moduliert Spermidin zelluläre Signalwege, die an Entzündungen und Immunreaktionen beteiligt sind.

Application

Spermidin wird wie folgt verwendet:
  • als Komponente von Chromosom-Isolationspuffer
  • als Komponente von Transkriptionspuffer
  • als Komponente von Homogenisierungspuffer

Biochem/physiol Actions

Hemmt die neuronale Stickoxidsynthase (nNOS). Bindet und fällt DNA aus; kann zur Aufreinigung von DNA-Bindungsproteinen verwendet werden. Stimuliert die T4-Polynukleotidkinaseaktivität.

  1. How should one provide inert gas atmosphere for storage after opening the provided vial? Do aliquots (diluted to 1M) need to be kept under inert gas as well? How long can one store said aliquots without impairng activity?

    1. After dispensing, briefly flush the vial headspace with dry nitrogen or argon and cap immediately. The headspace may be cleared by directing the inert gas across the opening of the vial or bottle. Alternatively, handling in an inert-gas enclosure, such as a glove box or Aldrich® AtmosBag (Z530212), is also suitable. Prepared 1 M solutions are stable for 3 to 6 months when stored refrigerated at 2 -8 °C and up to 12 months at - 20 °C, when stored in single use aliquots. Avoid repeated freeze/thaw cycles. While there is no specific guidance on clearing the headspace for prepared solutions, this additional measure would further ensure stability.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.     1. This product is considered a pure material rather than a solution. The concentration can be determined by dividing the density by the molecular weight and correcting for units. This gives 6.37M, which would then need to be multiplied by the batch-specific density for the batch in question, as the material is not 100% pure.

    1. Both items are from Buchs. Item 85558-1g comes in a narrow neck glass bottle, 5 ml, amber. Item 43816-10mL is a 15 ml plastic tube made of polypropylene (PP) and includes a sterile cap.

While considered stable, this item may be subject to degradation over time and is assigned a periodic retest date to confirm quality.
