Blasticidin S gehört zu der Aminoacylnukleosidklasse der Antibiotika. Es hemmt die Proteinsynthese in Bakterien und Eukaryoten.[1] Es weist außerdem fungizide Eigenschaften auf und verhindert Reisbräune.[2]

Anwendung
Blasticidin S wird als Selektionsmittel für umgewandelte Zellen, die die Resistenzgene bls, bsr oder BSD enthalten, verwendet. Blasticidin S wird verwendet, um HEK293-T-Zellen mit TLR-2-Konstrukten und HEK-D5-Zellen auszuwählen[3][4]. Es wird außerdem zur Untersuchung der Proteinsynthese auf Ebene der Peptidbindungsbildung verwendet.
Biochem./physiol. Wirkung
Blasticidin S hemmt die Hydrolyse von Peptidyl-tRNA, eingeleitet durch Freisetzungsfaktoren. Es verbessert die Bindung von tRNA an die große Untereinheit von Ribosomen, und hemmt in gewissem Maß die Bildung von Peptidbindungen.[1]
