General description

L-Malat: NAD⁺-Oxidoreduktase
Das L-Malat-Dehydrogenase-Enzym ist ein nukleares Genprodukt, das mit einem aminoterminalen Signalpeptid mit 24 Residuen synthetisiert wird. Dieses Peptid wird während der Translokation des Enzyms zur Mitochondrienmatrix proteolytisch gespalten.[1]

Application

Das Enzym L-Malat-Dehydrogenase aus dem Schweineherz wird zur Messung der Phosphoenolpyruvatcarboxykinase(PEPCK)-Aktivität verwendet. Das durch PEPCK erzeugte Oxalacetat wird mittels Oxidation von NADH zu Malat reduziert, das wiederum mit einem Spektrophotometer bei 340 nm gemessen wird. Das Enzym wurde ebenfalls zur Messung von Oxalacetat durch spektroskopische Messung der Reduktion von NADH bei 340 nm verwendet.[2]

Biochem/physiol Actions

Das Enzym L-Malat-Dehydrogenase aus dem Schweineherz katalysiert die Oxidation von L-Malat zu Oxalacetat. Das Enzym ist eine NAD-abhängige mitochondrische Dehydrogenase, die im Tricarbonsäurezyklus funktioniert. Es ist ein Bestandteil des Malat-Aspartat-Shuttles, das die Reduktionsäquivalente über die innere mitochondrische Membran in Form von Malat transportiert.[1]
