Beschreibung

General description

Synthetisiert aus Isoamylnitrit und Wasserstoffperoxid. Peroxynitrit (ONOO–;) ist ein starkes biologisches Oxidierungsmittel, das mit DNA, Membran-Phospholipiden, Sulfhydryl-Gruppen und Tyrosin reagiert. Es wird mit einer Vielzahl von pathophysiologischen Gegebenheiten in Verbindung gebracht, darunter ARDS (Acute Respiratory Distress Syndrome, Schocklunge), Arthritis, Atherosklerose, entzündliche Darmerkrankung, Reperfusion nach Ischämie, N-Methyl-4-phenyl-1,2,3,6-tetrahydropyridin (MPTP)-vermittelte Neurotoxizität und septischer Schock. Dieses Produkt zerfällt oder isomerisiert bei pH 8 oder darunter, die Halbwertszeit bei pH 7,5 beträgt ~ 1,5 s. ε;^302 nm = 1670^M-1cm^-1 in 0,1 MNaOH.

Preparation Note

Peroxynitrit ist löslich in wässrigen Puffern, die mit einem Inertgas vorgespült und auf 4 °;C vorgekühlt wurden.

Vor Hitze, Licht, Metallionen und Luft schützen. Peroxynitrit ist instabil; rasch auftauen, in Aliquote aufteilen, die Röhrchen mit Inertgas spülen und einfrieren (-70 °;C). Die Aktivität nimmt bei -20 °;C mit ~ 2 %/Tag ab.
