Beschreibung

General description

Eine Dibenzofuranylsulfanamidovalin-Verbindung, die als potenter, auf das aktive Zentrum von Zink abzielender, MMP-12-selektiver Inhibitor (IC₅₀ = 2 nM gegen menschliche MMP-12) mit viel schwächerer Aktivität gegen MMP-12 von Mäusen/Ratten/Schafen (IC₅₀ = 160, 320 und 22,3 nM) oder menschliche MMP-3/13/14 (IC₅₀ = 351, 120 und 1100 nM) wirkt und praktisch unwirksam gegenüber menschlichem Agg-1/2 (IC₅₀ >;5 µ;M), TACE (IC₅₀ >;25 µ;M) und MMP-1/7 (IC₅₀ >;6 µ;M) ist. MMP408 ist bei Mäusen oral wirksam und mildert nachweislich die durch rhMMP-12 ausgelöste Entzündungsreaktion der Lunge bei Mäusen in vivo um mehr als 50 % (5 mg/kg, p.o., b.i.d.).
