Beschreibung

General description

Natürlich vorkommendes Alkaloid. Potenter, selektiver und zellpermeabler Inhibitor von Proteinkinase C (IC₅₀ = 660 nmol/l). Wirkt auf die katalytische Domäne von PKC. Ein kompetitiver Inhibitor in Bezug auf den Phosphatakzeptor und ein nichtkompetitiver Inhibitor in Bezug auf ATP. Mehr als zehnmal wirksamer als H-7, HCl (Katalognr. 371955). Hemmt die Thromboxanbildung und den Phosphoinositidmetabolismus in Blutplättchen. Leitet auch die apoptotische DNA-Fragmentierung und den Zelltod in humanen promyelozytischen HL-60-Leukämiezellen ein.

Preparation Note

Für eine vollständige Solubilisierung kann ein Erwärmen auf 40–50 °C mit sanftem Rühren erforderlich sein. Unmittelbar vor Gebrauch mit Puffer weiter verdünnen.

Nach der Rekonstitution in Aliquote aufteilen und einfrieren (-20 °C). Stammlösungen sind bei -20 °C bis zu 1 Monat haltbar.
