General description
El tamoxifeno (TC), un modulador selectivo del receptor de estrógenos (SERM), se usa contra el cáncer de mama tanto en hombres como en mujeres. También actúa como profiláctico contra el cáncer de mama en mujeres.[1] Este inhibidor de la proteína cinasa C y factor antiangiogenético es un profármaco que es metabolizado a los metabolitos activos 4-hidroxitamoxifeno (4-OHT) y endoxifeno por las isoformas CYP2D6 y CYP3A4 del citocromo P450. En el cáncer de mama, la actividad represora génica del tamoxifeno contra el ERBB2 depende del PAX2. El TC bloquea la producción del factor de crecimiento endotelial vascular (VEGF) estimulada por estradiol en las células tumorales de mama. Los pacientes con tumores positivos para los receptores de estrógenos tienen más probabilidad de beneficiarse del tamoxifeno. Application
El tamoxifeno se ha utilizado:
- en el ensayo MTS (3-(4,5-dimetiltiazol-2-il)-5-(3-carboximetoxifenil)-2-(4-sulfofenil)-2H-tetrazolio) y la tinción con cristal violeta para evaluar la viabilidad celular[2] [3]
- para inducir la ablación de Dicer en ratones Dicer condicionales, estudiar los efectos de la deficiencia de Dicer sobre la diferenciación y la función de los adipocitos.[4]
- para estudiar sus efectos biológicos en la línea celular Jurkat resistente a los glucocorticoides (GC), derivada de la leucemia linfoblástica aguda del linaje T (T-ALL)[5]
- para estudiar los efectos fisiológicos del tratamiento con tamoxifeno sobre el área preóptica (POA) del hipotálamo mediados por el receptor de estrógenos alfa (ERα) en ratones[6]

Preparation Note
El tamoxifeno es soluble en cloroformo a 50 mg/ml y produce una disolución transparente de incolora a amarilla pálida. También es soluble en metanol, etanol, 2-propanol y propilénglicol. También puede prepararse disolución de reserva de tamoxifeno en DMSO 10 mM. Sin embargo, es prácticamente insoluble en agua (la solubilidad es <0,01 %, 20° C). Las disoluciones son sensibles a la luz UV, y las disoluciones de DMSO son estables cuando se conservan a -20° C en la oscuridad.

Hello, Thank you for your product. According to my protocol, I have to dissolve Tamoxifen in the corn oil at the final concentration of 40 mg/ml and feed my mice with that. However, it's insoluble. How can I resolve it? Thank you, Hossein

If I want to use 20mg of tamoxifen, could I only dissolve part of tamoxifen and then store the rest of the powder at refrigerator? Or do I have to dissolve all vial at once?

Tamoxifen is relatively stable. Any required amount can be weighed up for use as needed and the bottle resealed. The powder should be stable for at least two years when stored desiccated at 2-8C, and protected from light.

What is the solubility of tamoxifen, Product No. T5648?

Tamoxifen is tested for solubility in chloroform at a concentration of 50mg/mL. This yields a colorless to faint yellow solution. Tamoxifen is practically insoluble in water. It has been reported to be soluble in methanol, ethanol, 2-propanol and propylene glycol. This product is also reported to be soluble in DMSO at 10mM concentration. What is the stability of tamoxifen (e.g., Product No. T5648) once in solution?

Solutions of tamoxifen are sensitive to UV light. According to C.A. O'Brian et al., [Cancer Res. 45, 2462 (1985)], tamoxifen when dissolved in DMSO can be stored at -20°C in the dark. We generally recommend making the solution fresh right before each use.
