Application

SB-431542 was used to study the correlation of platelet-derived growth factor receptor B expression and TGF-β pathway activation in colorectal tumor cells.[1] It was used to study the role of TGF-β signaling in modulation of extracellular matrix[2] and myoblast differentiation.[3]

Biochem/physiol Actions

Potent and selective inhibitor of transforming growth factor-β superfamily type I activin receptor-like kinase (ALK) receptors.
SB-431542 inhibits the TGF-β-mediated activation of SMAD proteins, expression of collagen and fibronectin, cell proliferation and cell motility.[4][5][6] It does not inhibit kinases that are activated in response to serum or stress such as ERK, p38 or JNK.[5]

  1. Is it okay to store the original product and its aliquoted stock solutions not only at -20℃ specifically, but also in some temperature range around -20℃? Can I have certain temperature range for storage?

    1. The acceptable temperature range for products stored at -20°C is -10°C to -25°C. The stability of this product in solution has not been investigated. However, external sources indicate that stock solutions prepared in DMSO are stable for 6 months when stored at -20°C. The same acceptable range would apply.

  3. How long can I store the reconstituted compound in the freezer -20 degrees? i have one that was aliquoted 4 years ago. Is it still ok to use it ?

    1. It is recommended, following reconstitution, to aliquot and freeze -20°C. The stock solutions are stable for up to 6 months at -20°C. The quality of solutions stored frozen for extended periods has not been evaluated. The end user would have to determine suitability. Any color change, cloudiness, or particulate matter present upon thawing would be a visual indicator of degradation.
