General description

N-Nitrosodiethylamine belongs to the nitrosamines category of nitrogenous disinfection byproduct. It is also generated in chlorine based water treatment plants.

Application

N-Nitrosodiethylamine has been used to induce hepatocarcinogenesis in rat and mice.

Biochem/physiol Actions

N-Nitrosodiethylamine (NDMA) is carcinogenic in all animal species tested. The main target organs are the nasal cavity, trachea, lung, esophagus and liver. NDMA is acted upon by the cytochrome P450 system resulting in the formation of carcinogenic methyldiazonium ion. However microbes like Pseudomonas metabolize NDMA into N-nitromethylamine (NTMA) and formaldehyde.[1]

Cancer research has revealed that the classical model of carcinogenesis, a three step process consisting of initiation, promotion, and progression, is not complete.

DNA damage and repair mechanism is vital for maintaining DNA integrity. Damage to cellular DNA is involved in mutagenesis, the development of cancer among others.
