Unit Definition

1 U corresponds to the amount of enzyme which liberates 1 μmol glucose from carboxymethylcellulose per minute at pH 5.0 and 37°C; further enzyme activities: pectinase, xylanase, dextranase, laminarinase, amylase, protease, macerating and lytic activity; commonly used concentration: 1.0-2.5 %

Preparation Note

Preparation of protoplasts:
Place 0.2 g driselase in a conical flask, dissolve in 10 mL 4 M mannitol and adjust the pH with 0.1N HCl to 5.2. Suspend 1 g of cultured cells in the solution. When the enzyme is allowed to react with the cells for 1-2 hours while gently shaking the flask in a water bath at 30°C, most of the cells used as materials lose the cell walls and become spherical protoplasts.
