General description

Tyrosine-protein phosphatase non-receptor type 22 (EC:3.1.3.48; UniProt Q9Y2R2; also known as Hematopoietic cell protein-tyrosine phosphatase 70Z-PEP, Lymphoid phosphatase, Lymphoid-specific protein tyrosine phosphatase, Lyp, PEP, PEST-domain phosphatase, Protein tyrosine phosphatase non-receptor type 8) is encoded by the PTPN22 (also known as LYP, LYP1, LYP2, PTPN8) gene (Gene ID 26191) in human. PTPN22/Lyp is expressed in all hematopoietic cells and belongs to the PEST group of nonreceptor classical class I protein tyrosine phosphatases (PTPs). Lyp is a negative regulator of TCR signal transduction by targeting phosphorylated Lck (Y394), Fyn (Y427), and Zap-70, as well as phospho-sites on TCRζ, CD3ε, Vav, and Vcp. Lyp utilizes its proline-rich region 1 to interact with the SH3 domain of C-terminal Src kinase (Csk), resulting in enhanced inhibitory tuning of Src family kinases and TCR signal transduction. TRAF3 is also shown to bind the proline-rich region 1 of Lyp in myeloid cells and impact its turnover. Additional binding partners of Lyp include the adaptor molecule Grb2, the E3 ligase c-Cbl.

Specificity

Clone 3B3.1 is expected to react with spliced isoform 1 (LyP1), isoform 3, and isoform 4 (LyP3), but not isoform 2 (LyP2) or isoform 5. Reactivity toward isoform 6 (PTPN22.6) has not been tested.

Quality

Evaluated by Western Blotting in human burkitt′s lymphoma Ramos cell lysate.

Western Blotting Analysis: A 1:2,000 dilution of this antibody detected PTPN22 in 10 µg of human burkitt′s lymphoma Ramos cell lysate.

Target description

~92 kDa observed. 91.7 kDa (isoform 1; Lyp1), 85.1 kDa (isoform 3), 88.6 kDa (isoform 4; Lyp3) calculated. Uncharacterized band(s) may appear in some lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
