Mpro, 3CL 是病毒生命周期的关键酶。其活性产生对基因组复制和冠状病毒病毒粒子的产生至关重要的非结构蛋白:RNA 依赖性 RNA 聚合酶、解旋酶、核糖核酸酶和 3CLpro 本身,来自两类多蛋白(pp1a 和 pp1ab)。SARS-Cov2 病毒(SARS 冠状病毒)的成熟依赖于两种病毒蛋白酶对重叠的大型多蛋白 1a 和 1ab 的裂解:
  • Mpro(主蛋白酶)
  • PLpro(木瓜蛋白酶样蛋白酶)

SARS-CoV Mpro, 3CL 作为同型二聚体而存在,每个原聚体都有一个活性位点。Mpro 对 1ab 多蛋白的蛋白水解裂解发生在 11 个位点:1a 多蛋白内有 7 个位点,1ab 多蛋白内有 11 个位点。这导致 16 种病毒非结构蛋白的成熟。Mpro 蛋白酶形成功能性同源二聚体。Mpro 的 N 端和 C 端均对二聚体形成和酶功能至关重要。

  1. Dear Officer Please could you give me information on how to dissolve M-Pro Protease (SAE0172)? Which solvent to use? Also, what it is the optimal concentration for M-Pro to be stored? Thank you in advance

    1. It is suggested to reconstitute the product in water or in 10% aqueous glycerol solution at a protein concentration of 1-2 mg/mL. Aliquot the protein solution and store the solution aliquots at –20 °C. Avoid freeze-thaw cycles.
