生化/生理作用

使用神经氨酸酶去除细胞表面上糖蛋白的唾液酸残基已有多次报道。通常，实验步骤中会表明将神经氨酸酶在PBS中，37℃条件下孵育30分钟进行使用，然后用PBS洗涤几次。用浓度低得多的神经氨酸酶处理组织切片需要更长的温育时间：在pH 4.2-5的0.1M乙酸盐缓冲液中，对于1-4U/mL浓度，需要在37℃下孵育2-20小时。
利用神经氨酸酶裂解唾液酸基团已用来研究糖蛋白结构的抗体识别。与其他两种方法进行比较，使用神经氨酸酶可更好的对N-乙酰神经氨酸进行估测。
神经氨酸酶可用于从多种糖蛋白中切割末端N-乙酰神经氨酸（唾液酸）。 来自产气荚膜梭菌的酶可切割末端唾液酸残基，这些残基可通过α-2,3-、α-2,6-或α-2,8-与Gal、GlcNac、GalNAc、AcNeu、GlcNeu、寡糖、糖脂或糖蛋白进行连接。切割的相对速率按以下顺序逐渐降低：α-2-3>α-2-6。α-2-8。与来自A. ureafaciens的神经氨酸酶（Sigma N3642）（优先切割α-2-6连接的残基）相比，来自产气荚膜梭菌的神经氨酸酶可最有效地切割α-2-3连接的唾液酸残基。
