己糖激酶负责催化糖酵解的第一步。它导致葡萄糖和 ATP 形成葡萄糖-6-磷酸和 ADP。 葡萄糖-6-磷酸脱氢酶是磷酸戊糖途径中的关键酶。负责葡萄糖-6-磷酸和 NADP +(烟酰胺腺嘌呤二核苷酸磷酸)转化为 6-磷酸葡萄糖酸内酯和 NADPH。

單位定義

在+ 25°C和pH 7.6条件下,1个单位(U)HK将在1分钟内磷酸化1μmol的D-葡萄糖。
在+ 25℃和pH 7.6条件下,1单位(U)G6P-DH将在1分钟内氧化1μmol葡萄糖-6-磷酸。
在偶联试验中,每μmolD-葡萄糖磷酸化产生1μmolNADH。

体积活性:以葡萄糖和ATP为底物,+ 25℃时产生340u己糖激酶/ml。170 U葡萄糖-6-磷酸脱氢酶/ ml(+ 25°C,葡萄糖-6-磷酸为底物)

外觀

用 3.2 M 硫酸铵溶液(pH 值约为 6)混悬。己糖激酶与 G6P-DH 混合制备。HK:G6P-DH的蛋白质含量约为2:1。
