特异性

来自绵羊的多克隆抗体对地高辛配基和地高辛具有特异性，并且与其他类固醇（如人类雌激素和雄激素）没有交叉反应性。

应用

抗地高辛-POD（poly）Fab片段可用于检测洋地黄毒苷标记化合物，使用：
  • 整胚原位杂交[3]
  • 荧光原位杂交[4]
  • 酶联免疫吸附测定(ELISA)[5]
  • 斑点杂交
  • Southern印迹
  • 蛋白质印迹
  • 双重标记实验

与ELISA应用中的未聚合辣根过氧化物酶偶联抗地高辛-Fab片段相比，抗地高辛-POD（poly）Fab片段的信噪比高得多。当需要高灵敏度时，它特别有用。
抗地高辛-POD（poly），Fab片段尚未在免疫组织化学中进行评估。

制备说明

工作浓度：结合物的工作浓度取决于应用类型和底物。以下浓度可作为指导：
  • 斑点印迹：50至100mU/ml
  • ELISA：2至50mU/ml
  • 蛋白质印迹：50至100mU/ml
  • Southern印迹：50至100mU/ml

工作溶液: 100mM Tris-HCl，150mM NaCl，pH 7.5。
1％封闭试剂(w/v)，1至5％热灭活的胎牛血清(v/v)或绵羊正常血清（可用于减少非特异性结合）。

重悬

加入1ml双蒸水至终浓度为50U/ml。
