一般描述

百日咳毒素（PT）是百日咳杆菌分泌的蛋白外毒素和致病因子，是百日咳的病原体。[1]它通过由ptl基因编码的IV型分泌系统穿过细菌外膜进行运输，ptl基因位于编码毒素的ptx基因的下游。这种具有AB5结构的复杂多亚单位毒素是非细胞百日咳疫苗的关键成分。作为单组分类毒素它可以发挥保护作用。此外，由于其对G蛋白偶联受体途径的抑制作用，它被广泛应用于哺乳动物细胞信号传导研究。它抑制中性粒细胞募集和气道内流（airway influx），这是对细菌感染的常见炎症反应。[2]

应用

来自百日咳杆菌的百日咳毒素已被用作Gi蛋白抑制剂，用于研究百日咳毒素减少下丘脑阿丙鸦片黑素皮质素（POMC）神经元去极化反应的能力。[3]在百日咳毒素实验中，它还可用于阻断Gi，进行体内内耳和体外外毛细胞（HC）研究。[4]

在50 mM NaCl，10 mM磷酸钠缓冲液，pH 7.0中冻干。

重悬

在复溶后，冷藏（4°C）。为了在溶液中长期保存，请使用高离子强度缓冲液（例如100 mM磷酸钠，500 mM氯化钠，pH值7.0）复溶。贮备液在CHO细胞试验中可稳定至少6个月，在非细胞系统中可稳定1-2个月。
用500µl H₂O复溶。每次使用前，刮擦管壁并轻轻移取溶液混合，以获得均匀的混悬剂。
