General description

MHY1485, a small-molecule activator of the mammalian target of rapamycin (mTOR), exerts an inhibitory effect on the autophagic process by preventing the fusion between autophagosomes and lysosomes, resulting in the accumulation of LC3II protein and the enlargement of autophagosomes. It can also protect osteoblasts from dexamethasone-induced damage by inhibiting the mitochondrial death pathway provoked by dexamethasone.

Application

mTOR Activator, MHY1485 has been used to activate the mammalian target of rapamycin (mTOR) pathway in mice with Parkinson disease (PD) to reverse the effects of tumor necrosis factor α–induced protein 3 (TNFAIP3) on the mitigation of injury in mice and in SK-N-SH cells induced with 1-Methyl-4-phenylpyridinium (MPP+).

Autophagy is a major degradative process responsible for the disposal of cytoplasmic proteins and dysfunctional organelles via the lysosomal pathway. During the autophagic process, cells form double-membraned vesicles called autophagosomes that sequester disposable materials in the cytoplasm and finally fuse
