Purified bovine NF-H.

Application

Anti-Neurofilament H Antibody is an antibody against Neurofilament for use in IC, IH, IH(P) & WB.
Western blot: 1:1,000-1:10,000 using ECL. Recognizes a band of 200-220 kDa.

Immunohistochemistry: 1:1,000-1:10,000 on frozen or paraffin embedded tissue sections. Suggested fixative is 4% paraformaldehyde.

Immunocytochemistry: 1:1,000-1:10,000 on cells in tissue culture fixed for one minute in 3.7% formalin and one minute in -20°C methanol.

Optimal working dilutions must be determined by the end user.

Biochem/physiol Actions

Recognizes Neurofilament-Heavy (NF-H). The antibody stains sharply defined cytoplasmic filaments in neuronal cell bodies and especially axons.

  1. I am staining FFPE postmortem human spinal cord tissues. What does it mean if some motor neuron cell bodies have intense NF-H signal? Also should I interpret weak signals in the white and grey matter as background? Dendrites? Degenerating axons?

    1. AB5539 is the anti Anti-Neurofilament H (200 kDa) Antibody. I does stain the cytoplasmic filaments in neuronal cell bodies, and especially axons. In my opinion, most likely the weaker and stonger staining pattern is due to the NF antibody reacting to phosphorylated epitope in the axon. You may wish to review genral literature on this line, e.g. literature PMID: 3133402, PMID: 6416876,

  2. Does this antibody detect phosphorylated or non-phosphorylated NFH or both?

    1. This product is a polyclonal antibody that was made against biochemically isolated NF-H purified from bovine spinal cord. This preparation is dominated by axonal forms of NF-H which are heavily phosphorylated on the multiply repeated NF-H KSP type sequences, and this antibody reacts very strongly with these phosphorylated repeats. Reactivity with non-phosphorylated KSP sequences is orders of magnitude weaker. In most species there is some cross-reactivity with the phosphorylated KSP sequences found in the related neurofilament subunit NF-M which are similar but not identical to those of NF-H. The antibody recognizes phosphorylated NF-H strongly in all mammals tested to date.
