Clone 2L4 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Dopamine D1 receptor. It targets an epitope within 22 amino acids from the C-terminal cytoplasmic domain of the protein.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal cytoplasmic domain of human Dopamine D1 receptor.

Zastosowanie

Quality Control Testing

Evaluated by Western Blotting in SH-SY5Y cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Dopamine D1 receptor in SH-SY5Y cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Dopamine D1 receptor in human brain and rat brain lysates.

Affinity Binding Assay Analysis: The binding affinity of this antibody to Dopamine D1 Receptor peptide was at least one thousand-fold (1,000X) higher than that obtained with the non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Dopamine D1 receptor in A549 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Dopamine D1 receptor in human cerebral cortex tissue sections.

Dopamine D1 receptor (UniProt: P21728; also known as D(1A) dopamine receptor) is encoded by the DRD1 gene (Gene ID: 1812) in human. It belongs to the G-protein coupled receptor 1 family. Dopamine D1 receptor is composed of seven transmembrane domains and an extracellular N-terminus. It can form homo- or heterodimers with other GPCRs, which can influence its signaling and downstream effects. plays a critical role in dopaminergic neurotransmission. Upon activation by dopamine, the receptor initiates intracellular signaling pathways, including the stimulation of adenylyl cyclase and cyclic AMP production. Dopamine D1 receptor is primarily found in the central nervous system, particularly in brain regions associated with dopamine neurotransmission, such as the striatum and prefrontal cortex. Additionally, it is also present in other tissues to a lesser extent, including the heart, kidney, and adrenal gland. Dysregulation or dysfunction of the receptor has been associated with several neuropsychiatric disorders, including schizophrenia, bipolar disorder, and addiction. Variations in the gene encoding the receptor may influence the susceptibility to these conditions and affect individual responses to dopamine-related therapies. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Postać fizyczna

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Rekonstytucja

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Przechowywanie i stabilność

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
