Quality Control Testing

Evaluated by Western Blotting in HeLa cell nuclear extract.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TRIP12 in Human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound TRIP12 peptide with a KD of 7.3 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TRIP12 in U2OS cells.

E3 ubiquitin-protein ligase TRIP12 (UniProt: Q14669; also known as EC:2.3.2.26, E3 ubiquitin-protein ligase for Arf, HECT-type E3 ubiquitin transferase TRIP12, Thyroid receptor-interacting protein 12, TR-interacting protein 12, TRIP12) is encoded by the TRIP12 (also known as KIAA0045, ULF) gene (Gene ID: 9320) in human. TRIP12 is a nuclear protein that serves as an E3 ubiquitin-protein ligase involved in ubiquitin fusion degradation (UFD) pathway and regulation of DNA repair. It acts as a key regulator of DNA damage response by acting as a suppressor of RNF168, an E3 ubiquitin-protein ligase that promotes accumulation of  lysine 63-linked histone H2A and H2AX at DNA damage sites, thereby acting as a guard against excessive spreading of ubiquitinated chromatin at damaged chromosomes. TRIP12 is shown to be essential for cell viability and homozygous mutation that disrupts its ubiquitin ligase activity leads to embryonic lethality in murine models. In normal cells, TRIP12 is reported to mediate ubiquitination and degradation of isoform p19ARF/ARF of CDKN2A, a lysine-less tumor suppressor required for p53/TP53 activation under oncogenic stress. However, in cancer cells, p19ARF/ARF and TRIP12 are located in different cell compartments, which prevents their ubiquitination and degradation. Its expression is altered in several types of cancer, such as breast and pancreatic cancer and its higher expression is associated with poor prognosis in hepatocellular carcinoma.  Four isoforms of TRIP12 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Rekonstytucja

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
