General description

Trehalose, also known as mycose or tremalose, is a nonreducing disaccharide of glucose commonly found in fungi, bacteria, and invertebrates. Its presence in these organisms highlights its significance in various biological processes and potential applications in biochemical research and cell culture studies.

Application

D-(+)-Trehalose dihydrate has been used:

  • as a transcription factor EB (TFEB) to study its effects on an autophagy-lysosomal system in human silicosis alveolar macrophages[1]
  • as stabilizing excipient in spray-dried protein formulations[2]
  • as a cryoprotectant to study its effects on mouse testicular cell suspensions[3]

Biochem/physiol Actions

D-(+)-Trehalose shows anti-desiccant and cryopreservative activities. It acts as an osmolyte, and stress protectant and helps in the storage and transport of carbon. Trehalose is involved in the food, cosmetic, and pharmaceutical industries. It plays a vital role in the survival of certain insects and plants known as anhydrobionts during harsh environments such as high rates of dehydration.

Preparation Note

Prepared by ion exchange chromatography.

Questions

1–3 of 3 Questions
  1. I am planning to use this in an assay that may involve the amplification of fungal DNA via PCR. Since this is derived from S. cerevisiae, how can I be sure that it is DNA free?

    1 answer
    1. This product is not guaranteed to be DNA-free, as no specification exists for this. It is recommended to perform a sensitive PCR on the product. Specific primers can be designed to amplify S. cerevisiae DNA, and a PCR reaction can be conducted using this product as the template. If no amplification occurs, it suggests that the reagent is DNA-free.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf
