General description

Trehalose is a non-reducing disaccharide, ubiquitously present in nature. It is used as a food preservative and also to retain enzymes, vaccines, cells and many other molecules. Trehalose is present as hemolymph sugar among the invertebrates.

Application

D-(+)-Trehalose dihydrate has been used in a slow-freezing method for freezing rabbit oocytes. It has also been used in solid-surface vitrification (SSV) of in vitro-matured oocytes. It is suitable for use as a cryoprotectant in a variety of cell freezing media.

Biochem/physiol Actions

Trehalose is essential for embryogenesis, carbon metabolism, photosynthesis and flowering in plants. It predominantly exists among the species that supports anhydrobiosis (life without water). It serves as a carbohydrate reserve in microorganisms and protects them from adverse conditions, such as heat, cold, dehydration, desiccation and oxidation. Trehalose acts as a chemical chaperone affecting protein folding. It is known to prevent amyloid generation and accummulation of β-amyloid in vitro. A similar effect of trehalose has been observed with huntingtin protein aggregation. Thus, it might serve as a protective agent in Alzheimer′s disease and Huntington′s disease.

  1. What is the average particle size in microns or particle mesh size for this product, please? This is important for my research work.

  2. We would like to dissolve trehalose in water in bulk and store it. Does dissolved trehalose loose its stability after an extended storage time? Which storage temperature would you recommend?

    1. The solution stability and storage of this product has not been tested, however other commercial sources offer 1M Trehalose solutions that are stored at ambient temperature or refrigerated (filter-sterilized). Trehalose is added to several of less stable protein preparations as a carrier and/or cryoprotectant. An example would be EGFR, E2645. This lyophilized product contains 10% Trehalose. Upon reconstitution the product can be stored for up to 1 year at -70 deg C. Given the information above, Trehalose may be quite stable in solution.

    1. Do not autoclave. It is best to sterile filter solutions.

    1. Yes, it should work as a substitute for protein in a freezing media.  It is commonly used at a final concentration of 5% (50 mg/mL).  Alternatively, one could use methylcellulose.
