General description

Pyrophosphate (PPi) is produced by a number of biochemical reactions such as ATP hydrolysis, DNA and RNA polymerizations, cyclic AMP formation, and the formation of fatty acid-coenzyme A esters.

Application

The Pyrophosphate Assay Kit provides a simple and direct flourescent microplate procedure for measuring pyrophosphate in a variety of samples.

Biochem/physiol Actions

The pyrophosphate concentration of a sample is determined by the use of a unique fluorogenic pyrophosphate sensor in which the presence of pyrophosphate results in the production of a fluorescent product (λex =370/λem = 470 nm) proportional to the pyrophosphate present. This assay is simpler and more robust than traditional enzyme-based methods and is ideal for screening enzyme activity or enzyme inhibitors.

Questions

1–2 of 2 Questions
  1. Can this kit be used to quantify pyrophosphate in cell extracts? If so, are there any recommendations for sample preparation?

    1 answer
    1. The kit has not been validated for use with cell lysates. No supporting data are currently available for this sample type. The end user would have to determine suitability. Please refer to the link below to review the product datasheet:

  2. The product info sheet recommends 10-30min incubation after probe addition. Is this kit therefore amenable to continuous read formats or is signal development slow? Also, is there a recommended quenching method for this kit?

    1 answer
    1. This kit functions using an endpoint determination. Fluorescence intensity should be measured a single time between 10 and 30 minutes after the addition of the master reaction mix. The reaction signal remains stable for 2 hours. No recommended quenching method is available.
