For assay purposes, this product is reconstituted at 1 mg/mL in 0.9% NaCl. Stock solutons are used immediately. For extended storage, the higher concetration of 5 mg/mL may assure maximum recovery. Lectins have a tendency to stick to surfaces. When stored at lower concentrations, any loss due to surface adhesion is more impactful. Note that stock solutions should be well mixed after thawing to assure maximum potency.

Lectins are carbohydrate-binding proteins and are highly specific for sugar moieties. Pokeweed lectin (PL) is a lectin specific for N-acetylglucosamine-containing saccharides that stimulate peripheral lymphocytes to undergo mitosis by binding to their cell surfaces. Four types of lectins are isolated from the roots of pokeweed (Phytolacca americana). They contain homologous domains but have different molecular sizes and biological properties.
PWM is extracted from pokeweed roots. This protein has hemagglutinating, leukoagglutinating, and mitogenic properties. PWM is not blood group specific, but has affinity for N-acetyl-β-D-glucosamine oligomers.

Mitogenic activity was determined by BrdU incorporation in HeLa cell cultures.

I want to use this for B cell stimulation. Is it better to reconstitute in 1 mL (5mg/mL) or 5mL (1mg/mL) PBS for this? I will use this to stimulate PBMCs at a concentration of 3 ug/mL (as recommended in the previous post).

This product can be dissolved in PBS and used at up to 3 ug/ml for mitogenic effects on human peripheral blood mononuclear cells. Any concentration higher than 3 ug/ml could be toxic to these cells.
