Janus kinase 2(JAK2) mapped on human chromosome 9p24, encodes a tyrosine kinase. The encoded protein is a member of the Jak family of protein. JAK2 is ubiquitously expressed. This protein contains a catalytic domain and protein–protein interaction domain.

JAK2 mutations promote primary resistance to PD-1 (anti-programmed death protein 1) blockade therapy. In addition, mutations in the gene has also been observed in polycythaemia vera patients.
