一般描述

免疫球蛋白G(IgG)是免疫球蛋白家族的成员,并且在恒定(C)区中包含γ(γ)重链。IgG的单体结构包含两条相同的重链和两条相同的轻链,分子量分别为50kDa和25kDa。该抗体的一级结构还包含连接两条重链、连接重链和轻链、并位于链内的二硫键。IgG进一步分为四类具有不同重链的IgG1、IgG2、IgG3和IgG4,分别命名为γ1、γ2、γ3和γ4。

特异性

通过免疫电泳 (IEP) 与正常小鼠血清和小鼠 IgG 比较,确定抗血清对小鼠 IgG 具有免疫特异性。通过 Ouchterlony 双扩散 (ODD),产物与小鼠 IgG、小鼠 IgA 和 IgM 反应。

免疫原

使用纯化的小鼠 IgG 作为免疫原,在家兔中产生抗小鼠 IgG。纯化整个抗血清以提供抗血清的 IgG 部分。

应用

在兔中产生的抗小鼠 IgG(全分子)抗体在免疫印迹、免疫沉淀 和免疫组织化学中用作二抗。

生化/生理作用

IgG抗体亚型是免疫系统中最丰富的血清免疫球蛋白。它由B细胞分泌,存在于血液和细胞外液中,可防止细菌、真菌和病毒引起的感染。母体IgG可通过胎盘转移至胎儿,胎盘对新生儿免疫防御感染至关重要。

外形

0.01 M 磷酸盐缓冲液 (pH 7.4) 中的溶液,含 15 mM 叠氮化钠作为防腐剂

储存及稳定性

需要连续使用时,可在2-8℃下储存一个月。对于更长期的储存,溶液可以冷冻成工作等分试样。不建议重复冷冻和解冻,或储存在“无霜”冰箱中。如果长期储存时出现轻微混浊,使用前通过离心澄清溶液。
