一般描述

FastStart Universal SYBR® Green预混液(Rox)是一种用于qPCR和RT-qPCR的即用型热启动反应混合物,适用于所有需要用ROX标准化的实时荧光定量PCR系统。

SYBR® Green I是一种DNA双链特异性染料。在DNA合成的每个阶段,包含在反应液中的SYBR® Green I染料可与扩增的PCR产物相结合。扩增子可通过荧光检出。

热启动实验方案经证可大幅提升特异性、灵敏度及PCR得率。在FastStart Taq DNA聚合酶的某些氨基酸残基上的热不稳定性封闭基团使得修改后的酶在室温(+15至+25°C)没有活性。因此,在引物可以非特异性结合的阶段并没有发生延伸。FastStart Taq DNA聚合酶可在高温(如在+95°C的预孵育步骤)下通过将封闭基团去除而被激活。

应用

FastStart Universal SYBR® Green预混液(Rox)可用于任何DNA或cDNA靶标(包括高GC或AT含量靶标)的扩增和检测。
将预混液与Transcriptor First Strand cDNA合成试剂盒(Roche)结合以在两步法qRT-PCR中实现极佳的结果。
FastStart Universal SYBR® Green预混液(Rox)已用于qRT-PCR[1][2][3][4][5]和RT-PCR。[6][7][8][9]

特点和优势

最大限度减低非特异性扩增产物的形成。

  • 避免对qPCR结果的过度预计。
消除可增加量化的SYBR Green I结合量的非特异性扩增产物和引物二聚体。
  • 对一系列的DNA或cDNA靶标进行扩增和检测。
扩增最长达500 bp的片段,包括高GC或高AT的。

  • 在qPCR制备中节省时间和工作。
免去了降成分混合、优化MaCl2浓度或进行其他耗时优化优化步骤的需要。

  • 防止来自残留污染的假阳性结果。
使用这种含有dUTP的预混液配合尿嘧啶-DNA糖基化酶对来自之前PCR反应的残留DNA污染进行清除。

组分

2x浓度的FastStart Universal SYBR Green Master (Rox)预混液含有FastStart Taq DNA聚合酶、反应缓冲液、核苷酸(dATP, dCTP, dGTP, dUTP)、SYBR Green I以及参考染料。

质量

功能测试:每个批次都采用三种类型模板对在qPCR中的性能进行测试:高GC模板、高AT模板及长片段模板(约440 bp)。
