转化生长因子β(TGFβ)系统属于细胞因子超家族和信号通路成员，在动物界进化上高度保守。TGFβ参与免疫细胞发育、血管生成、伤口愈合、胚胎发育和骨发育过程，在免疫应答、心脏病和肿瘤中起重要作用。正常状态的TGFβ是少数几种能够抑制细胞生长的蛋白，通过使有丝分裂停滞在G1期，促进细胞分化和凋亡。但是，TGFβ信号通路的癌变会使肿瘤细胞能够抵抗TGFβ的正常发挥作用，导致失去调控的增殖发生。初步研究表明，VE-钙粘蛋白可能增强突变的TGFβ信号通路。

MILLIPLEX® TGFβ1单重磁珠检测试剂盒是单因子分析试剂盒，用于定量分析血清、血浆、组织/细胞裂解物和培养上清液中的TGFβ
解决方案。

Luminex® xMAP®平台使用磁珠免疫分析格式，以实现理想的速度和灵敏度，同时定量多种分析物，从而显著提高生产力，同时节省宝贵的样品量。

特異性

除了人类TGFβ1，该试剂盒还可以检测下列动物血清中的TGFβ1：马、兔、小鼠、大鼠、小型猪、犬、仓鼠、豚鼠、恒河猴和食蟹猴。但是，准确的交叉反应量并未测定。

應用

  • 分析物：TGFβ1
  • 建议种属：除了人类TGFβ1，该试剂盒还可以检测下列动物血清中的TGFβ1：马、兔、小鼠、大鼠、小型猪、犬、仓鼠、豚鼠、恒河猴和食蟹猴。但是，准确的交叉反应量并未测定。
  • 建议的样本类型：血清、血浆、细胞/组织培养物上清液和裂解物
  • 注：本品需要采用提供的试剂和方法对样品进行酸化处理。
  • 建议的样本稀释方法：血清和血浆样本，在酸化处理后1:30稀释，每孔加样25 μL；细胞/组织培养样本，需要在酸化前稀释（详见说明书）。
  • 检测耗时：在2-8℃下过夜（16-18小时）或在室温（20-25℃）下2小时。
