胚胎干(ES)细胞是来自胚胎植入前哺乳动物胚胎内部细胞团(ICM)的全能细胞,能够在体外无限、未分化增殖[1,2,3]。 未分化的鼠类ES细胞可以在含有细胞因子、白血病抑制因子(LIF)或Millipore专有的ES细胞培养试剂ESGRO的培养基中实现长时间的体外维持′ [4,5]。 ES细胞的未分化状态表现为碱性磷酸酶(AP)的高水平表达[6]、包括SSEA和TRA抗原的表面标志物以及转录因子Oct-4的表达。

Millipore′碱性磷酸酶检测试剂盒(货号SCR004)是一种特异性和灵敏的工具,可通过确定AP活性来对ES细胞分化进行表型评估。

可从Millipore单独购买单克隆抗体TRA-2-49(产品货号MAB4349)和TRA-2-54(产品货号MAB4354),其可用于检测ES细胞表达的肝/骨/肾AP同工酶。 当用于流式细胞术时,这些试剂可以定量评估ES细胞对AP表达的影响[7]。

成分

快速红色紫罗兰色溶液(0.8 g/L储存液)(产品号90239)。两个15 mL瓶。

在AMPD缓冲液(pH 9.5;2 mol/L)中的Napthol AS-BI磷酸盐溶液(4 mg/mL)(产品号90234)。一个15 mL瓶。

儲存和穩定性
