一般說明

酪氨酸羟化酶在肾上腺素能神经元的生理中起重要作用。它是从酪氨酸生物合成儿茶酚胺多巴胺和去甲肾上腺素的第一种酶和限速酶。因此,TH是一种有用的多巴胺能和去甲肾上腺素能神经元标志物。TH酶活性维持需要亚铁离子作为辅因子,且人们普遍认为其受到磷酸化作用调控。目前至少鉴定出了四种因可变剪接而产生的人TH异构体。

特異性

可识别调节性N末端外部的表位可通过Western blot识别一个约59-61 kDa的蛋白。在Western blot检测时不要与以下酶发生反应:多巴胺-β-羟化酶、苯丙氨酸羟化酶、色氨酸羟化酶、脱氢蝶呤还原酶、Sepaapterin还原酶或苯乙醇胺-N-甲基转移酶(PNMT)。

應用

使用该抗-酪氨酸氢化酶抗体进行酪氨酸氢化酶检测,克隆LNC1已经验证可用于IH、IH(P)、IP &WB并有超过85篇的产品引文。
免疫组化(石蜡):
之前一个批次的1:200-1:400稀释液被用于IH。4% PFA固定的冷冻切片;4% PFA固定的石蜡切片1:100(Barrachina, M. et al., 2003)。针对石蜡切片,Barrachina曾报道过采用柠檬酸微波微波抗原回收成功进行过染色,但其他方法也应该可以使用。

免疫沉淀:
之前一个批次的该抗体被用于IP(Perez, 2002)。

终端用户需要对最佳的工作稀释比和实验方法进行确定。
