掺入DNA的溴脱氧尿苷(BrdU)的免疫细胞化学检测是研究正常细胞和赘生性细胞的细胞动力学的有力工具。用胸苷类似物BrdU体外或体内标记肿瘤细胞,然后用特异性抗BrdU单克隆抗体检测掺入的BrdU是定量DNA合成程度的准确而全面的方法。BrdU被掺入S期细胞的新合成DNA中,因此可以提供S期细胞比例的估计值。还可以通过双变量BrdU/DNA流式细胞术分析获得动态增殖信息(例如S期转运速率和潜在的倍增时间)。

质量
通过免疫细胞化学在HeLa细胞上评价了代表性批次。代表性批次的1:2-1:4稀释液在用95%乙醇,5%冰醋酸固定的HeLa细胞中显示出抗BrdU的阳性免疫染色。使用DNA复制检测试剂盒(目录号17-316)检测该批次。
