Interleukin-8 or CXCL8 (IL-8) is a proinflammatory chemokine that belongs to the C-X-C family. Monocytes and macrophages are the primary sources of IL-8 but it is also secreted by most nucleated cells. IL-8 binds to two G protein-coupled receptors, CXCR1 and CXCR2 and leads to the downstream activation of PI3K, MAPK, PKC, STAT3 and FAK pathways. IL-8 is responsible for the recruitment monocytes and neutrophils that results in a prolonged acute inflammation. The expression of IL-8 is influenced by inflammatory signals, chemical and environmental stress and hormones. Deregulation of IL-8 signaling has been observed in atherosclerosis, cardiovascular disease and solid tumors of bladder, prostate, ovary, glial cells and pancreas. Monoclonal Anti-Interleukin-8 antibody recognizes human IL-8. By immunoblotting, the antibody shows 100% cross reactivity with recombinant porcine IL-8, and no cross reactivity with recombinant rat CINC-2α.

Monoclonal Anti-Interleukin-8 antibody may be used for neutralization assays; ND50 is 0.08-0.4 μg/ml. The antibody may be used for immunoblotting at a working concentration of 1-2 μg/ml. For ELISA a working concentration of 0.5 μg/ml may be used. Antibody concentration of 25 μg/ml may be used for immunocytochemistry using stimulated human peripheral blood mononuclear cells. For flow cytometry, 10 μL of 25 μg/ml stock antibody solution may be used for 1-5 X 105 cells.
