General description
アンジオテンシン変換酵素（ACE）は、その活性部位に亜鉛結合モチーフHEXXHを持つ、体性および精巣のアイソフォームとして存在するジペプチジルカルボキシペプチダーゼです。ACEは、レニン-アンジオテンシン系を介して血圧を調節します。ACEは、アンジオテンシンIを重要な血管収縮剤であるアンジオテンシンIIに変換し、血管拡張剤であるブラジキニンを阻害することによって、血圧を上昇します。ACEの阻害は、高血圧の標的治療戦略です。カプトプリル、エナラプリル、およびリシノプリルを含む、幾つかのACE合成阻害ペプチドが臨床で使用されています。現在、天然食品資源由来の阻害ペプチドや、植物由来のフェノール化合物によるACEの阻害が研究されています。ACEは、精子膜のグリコシルホスファチジルイノシトール（GPI）に結合したタンパク質を遊離させることによって、受精において重要な役割を果たします。
Application
ウサギ肺由来アンジオテンシン変換酵素は以下のことに使用されています：
- 高速液体クロマトグラフィー（HPLC）を用いた、卵白加水分解物のACE活性に対する阻害効果の測定
- 基質としてヒップリル-L-ヒスチジル-L-ロイシン（HHL）を用いた、逆相-HPLC（RP-HPLC）3およびHPLCそれぞれによる、ライチ果皮および調理したトリ胸肉のACE阻害活性の測定
- HeLaやHEK293などの幾つかの細胞株におけるin vitroでの、およびマウス精子におけるin vivoでの、GPIアンカータンパク質の遊離。
Biochem/physiol Actions
作用を受けやすい基質(アンジオテンシン I、ブラジキニンなど)からC末端ジペプチドを除去します。
