p38ファミリーとは、JNK/SAPKファミリーのサブファミリーであるSAPK2/3キナーゼ（ストレス活性化タンパク質キナーゼ）を指します。p38/SAPKキナーゼは、ストレス（最も重要なのは、炎症性サイトカイン、放射線照射、およびアニソマイシンや亜ヒ酸塩などの特定の毒素など）によっても活性化されます。SAPKは、炎症、癌、および神経変性疾患を含む多くの病的状態に関与しています。他のMAPKファミリーメンバー（ERK、JNK、p38MAPK、ERK5）と同様、p38は、Thr-X-Tyr二重リン酸化モチーフ（p38ではXはGly）のスレオニン残基とチロシン残基の両方のリン酸化を介して活性化されます。リン酸化されて活性化されると、続いて細胞質と核の両方で下流の標的をリン酸化します。p38の主な標的の一部は、キナーゼMAPKAPK2と、そのリン酸化を介して活性化される転写因子ATF2です。
