Applicazioni
Il siero umano AB è usato in ingegneria dei tessuti, trapianto e applicazioni di terapia cellulare per l′espansione di cellule staminali mesenchimali (MSC) da tessuto adiposo o cellule mesenchimali stromali da midollo osseo umano; per la generazione di innesti di staminali epiteliali limbali standardizzate e trapianto; per espansione ex-vivo di cellule NK da sangue periferico in mezzo di espansione di cellule staminali ematopoietico; e per migliorare la tecnologia delle isole in coltura pretrapianto.
Il siero umano è stato impiegato: come componente del tampone fosfato salino con detergente Tween® (PBST) in un saggio di immunoassorbimento legato a enzima con anticorpo ponte ELISA (bridging ELISA), come campione biologico di controllo per la valutazione dei risultati di un saggio ELISA basato su carta su campioni biologici complessi, come diluente per creare una serie di diluizioni del campione di riferimento per la generazione di una curva standard per il NISTmAB aggiunto come spike nel siero, oltre che per il campione MM2232 di paziente con mieloma multiplo (MM)
Azioni biochim/fisiol
Il siero umano AB in genere si prepara lasciando coagulare il sangue intero naturalmente, ottenendo così quello che si chiama siero “off-the-clot (OTC). Il siero umano ottenuto dal sangue intero coagulato è preferibile in quanto contiene in genere livelli più alti ed una più ampia varietà di fattori di crescita, per una crescita delle cellule ottimale. Il siero umano da donatori con gruppo sanguigno AB è privo di anticorpi specifici per gli antigeni dei gruppi sanguigni A e B. Ciò lo rende un siero “donatore universale” indicato per diverse tipi di colture cellulari, tra cui le linee cellulari staminali umane.
Confezionamento
A causa della ridotta disponibilità delle bottiglie usate per confezionare questo prodotto, il confezionamento può cambiare da un lotto ad un altro. La presenza o la mancanza di una pellicola termoretraibile attorno al tappo della bottiglia non ha influenza sull′integrità né sulla sterilità del prodotto.
Attenzione
Ciascun donatore è testato e trovato negativo a epatite B & C e non reattivo secondo ELISA all′anticorpo del Virus di Immunodeficienza Umana (HIV). Tuttavia prodotti di origine umana dovrebbero essere considerati potenzialmente infettivi e trattati di conseguenza.

Each donor is tested for and found non-reactive to Hepatitis B & C and non-reactive to Human Immunodeficiency Virus (HIV) antibody by ELISA. HSV is not on the list of agents that are tested. Nevertheless, products of human origin should be considered potentially infectious and handled accordingly. If this is "serum" derived from "plasma" the plasma would have contained anti-coagulant. To turn the plasma into serum it has to be clotted. Has this product been clotted to remove the fibrin? Or is it still in fact plasma?

The product has been clotted. Blood is collected and treated with citrate (anticoagulant) which inhibits clotting initially. The blood is then centrifuged. When the spin is complete, layers will appear which allow a differentiation between the cells and the plasma. The plasma is removed and calcium is added to activate the clotting cascade. The preparation is then centrifuged again and the result is a large clot which is pelleted to the bottom of the tube with a serum layer on top.

Human Serum products H4522 and H6914 may not be suitable for complement assays when stored at -20°C due to potential degradation of complement activity. For complement assays, alternatives like S1764 (Complement sera human) and C9473 (Complement Serum Standard human) are recommended. Human AB serum is added as a supplement to culture media at a concentration of 2-10% to provide a mixture of nutritional, hormonal, growth, and attachment factors. The serum also acts as a buffer to the cell culture system against a variety of disruptions to cell growth and toxic effects such as pH change, proteolytic activity, or the presence of heavy metals or endotoxin.

According to the supplier:
Each donor unit used in the preparation of this material was tested and found negative for HBsAG, HBV-NAT, HIV-1/2, HCV, HCV-NAT, HIV-1 NAT, and Syphilis per FDA regulations. The human plasma was collected from male donors' blood type AB in FDA licensed commercial donor establishments located in the USA in accordance with ethical standards. Each donor signed an informed consent form prior to collection, which is traceable by the unique identifiable number to maintain anonymity. A quality assurance system is in place to ensure traceability to individual donor. Documentation to this effect is available upon request.Utile?

Could you please confirm if the serum contains normal levels of human complement components?

This product has not been treated to remove or inactivate complement components, however no testing has been performed to confirm these levels. It is a pooled product derived from 50 - 70 donors per lot. Any active complement be expected to meet 'normal levels.
