General description

ANTI-FLAG, l′anticorpo policlonale per affinità anti-FLAG di coniglio riconosce l′epitopo FLAG posto sulle proteine di fusione FLAG. Questo anticorpo reagisce con le proteine di fusione FLAG N-terminali, N-terminali-met e C-terminali.
L′epitopo FLAG è una proteina costituita da otto aminoacidi con un sito di scissione per l′enterochinasi.

Application

L′anticorpo riconosce l′epitopo FLAG sulle proteine di fusione taggate con FLAG in posizione N-terminale o C-terminale applicando i saggi dot blot, immunoblot, di immunoprecipitazione e immunocitochimici.

Biochem/physiol Actions

L′epitopo FLAG ha un ruolo cruciale nella purificazione per immunoaffinità delle proteine di fusione.

Physical form

Soluzione in tampone fosfato salino 0,01 M, pH 7,4, contenente l′1% di albumina da siero bovino e 15 mM di sodio azide.

Preparation Note

Purificato con cromatografia di affinità su una colonna contenente il peptide immunizzante.

  1. Can this AB (F7425) used for mouse 30um thick section. mouse will be perfused with 4% PFA and than cryopreserved. thanks

    1. This product has been recommended for use in immunofluorescence and indirect immunofluorescence at 5-10 μg/mL using 293T cells transfected with a plasmid encoding FLAG-JNK. It has not been tested for use in IHC applications. The end-user will need to optimize the assay. Please see the link below to review the product datasheet:

      In general, antibodies work well on 5-15 uM sections, for 30 uM sections, blocking /permeabilization steps and antibody dilution, the incubation time will need to be optimized by end user.     1. For a Western blot from a PAGE run (preferably) under denaturing conditions, a 1:400 dilution of this antibody should be used.  A 1-10 μg/ml concentration should be used when using thie antibody in an ELISA application.

  9. How to get rid of background when using Product F7465, ANTI-FLAG® from rabbit,?

    1. To reduce non-specific background on tissue staining, a small amount of Triton X-100 (Product No. X100 or T8787), can be added to the wash buffers and antibody incubation buffers. Try a Triton X-100 concentration of 0.1% to start. Tween 20 (Product No. P5927) can also be used.
