Descrizione generale

Falkor is expressed at different levels in various tissues. It is localized in the nuclei of cells and encoded by 2Kb mRNA. Falkor is ubiquitously expressed in various mouse tissue with high level expression in testis and the lowest expression in spleen and skeletal muscle.
Monoclonal Anti-Falkor/PHD1 (mouse IgG2a isotype) is derived from the FLK-49 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice. The PHD1 gene is mapped to human chromosome 19q13.2and exists in three isoforms.

Specificità

Monoclonal Anti-Falkor/PHD1 recognizes mouse Falkor/PHD1 (approx. 47 kDa).

Immunogeno

synthetic peptide corresponding to amino acids 2-20 of falkor/PHD1.

Applicazioni

Monoclonal Anti-Falkor/PHD1 antibody is suitable for microarray and western blot at a concentration of 0.5μg/mL using total cell extracts from 293T cells transfected with Falkor-myc tagged.
Monoclonal Anti-Falkor/PHD1 antibody produced in mouse may be used in enzyme-linked immunosorbent assay (ELISA) and immunoblotting.

Azioni biochim/fisiol

EGLN2 (Egl nine homolog 2), also known as prolyl hydroxylase domain-containing protein 1 (PHD1), functions as an oxygen sensor in cells and mediates oxygen homeostasis. It inhibits nuclear factor kappa-light-chain-enhancer of activated B-cells (NF-κB) signaling. Polymorphisms in the EGLN2 is implicated in various colorectal, hepatocellular and non-small cell lung cancer.
Falkor, a novel cell growth regulator, is speculated that the falkor protein is associated with suppression of cell growth and comprises of several putative phosphorylation sites for proteins such as protein kinase C and casein kinase II. Falkor has a significant homology to the C.elegans EGL-9. The human homolog of Falkor and EGL-9 is PHD1 (prolyl hydroxylase 1) which is involved in the regulation of HIF-1α. PHD1 function as dioxygenases that regulate HIF-1α by prolyl hydroxylation.

Stato fisico

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Stoccaggio e stabilità

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
