Application

La DNasi I è usata per tagliare il DNA come primo stadio dell′incorporazione di basi marcate nel DNA. L′enzima Sigma è stato utilizzato per trattare tessuto cerebrale di ratto. Questo studio ha dimostrato che la crescita degli assoni sugli astrociti non è inibita dagli oligodendrociti. In un altro studio, campioni fissati e scongelati di E. coli sono stati digeriti con DNasi Sigma insieme ad altri enzimi. La digestione è stata condotta prima della permeabilizzazione e colorazione degli acidi nucleici.
Desossiribonucleasi I da pancreas bovino è stato impiegata in uno studio per investigare l′analisi bidimensionale di zimogrammi delle nucleasi in Bacillus subtilis. Desossiribonucleasi I da pancreas bovino è stata usata in uno studio per investigare gli effetti di farmaci che si legano ai solchi maggiori e minori e intercalatori nella associazione al DNA di proteine legate al solco minore RecA e desossiribonucleasi I.
Usato per rimuovere DNA da campioni di proteine.

Biochem/physiol Actions

La DNasi I è un′endonucleasi che agisce sui legami fosfodiesterici adiacenti alle pirimidine per produrre polinucleotidi con fosfati 5′-terminali. In presenza di Mg2+, la DNasi taglia ciascun filamento di DNA indipendentemente e i siti di clivaggio sono casuali. Invece, in presenza di Mn2+ i due filamenti di DNA vengono tagliati approssimativamente alla stessa altezza. Il pH ottimale è fra 7 e 8. I cationi bivalenti come Mn2+, Ca2+, Co2+, e Zn2+ sono attivatori dell′enzima. Una concentrazione di Ca2+ 5 mM stabilizza l′enzima contro la digestione proteolitica. La DNasi I da pancreas bovino consiste di quattro componenti, A, B, C, e D, distinguibili cromatograficamente, i primi tre in rapporto molare 4:1:1. La forma D è presente solo in piccola quantità. 2-Mercaptoetanolo, chelanti, sodio dodecilsolfato (SDS) e actina sono noti inibitori dell′attività enzimatica.

Preparation Note

Una soluzione di DNAsi I 10 mg/mL in NaCl 0,15 M può perdere <10% della sua attività quando conservata per una settimana in aliquote a −20 °C. Le stesse soluzioni conservate a 2-8 °C possono perdere circa il 20% dell′attività. Rimane attiva fino a cinque ore a 60 °C a pH fra 5 e 7 e perde la sua attività in <10 minuti a 68 °C. L′attività decade del 6% all′ora in tampone acetato (a pH 5,0) e tris (a pH 7,2) alla concentrazione di 1 mg/mL.

  1. How many milligrams are in one vial of D5025-150KU?

    1. This product is assigned a minimum activity specification of 2000 units per milligram protein and a minimum protein specification of 80%. At the minimum value for each specification, the 150,000 unit package size would contain 93.75 mg of powder using the following calculation:
      Units/(activity x %protein) = mass or 150,000/(2000 x 0.80) = 93.75 mg
      The activity and protein content are lot-specific and reported in the product Certificate of Analysis. As the activity increases the mass per package size decreases. Please see the link below to review a sample or lot-specific Certificate:

  2. Good afternoon, if the stock concentration of 10mg/ml may loose 10% of its activity after one week, I was wondering if it is okey regarding stability to keep 500ug/ml aliquots at -20ºC for several weeks.

    1. As cited in the product listing and datasheet, this product in solution shows an approximate loss of activity of 10% when stored in aliquots at −20 °C for 1 week. Studies at lower dilutions have not been performed. Typically, solutions stored at higher concentrations are the most stable.

  3. Is Kunitz unit same as unit here? I have seen some papers and they have U/ml for DNase. How can i conver it?

    1. Product D5025, Deoxyribonuclease I is given in Kunitz units. This product is a powder. When reconstituting the product with a solution, for example 15,000 Kunitz units in 1 ml of 0.15 M NaCl the end-user will have 15,000 Kunitz units/ml.

    1. Solutions of DNAse I (10 mg/ml) in 0.15 M NaCl may lose 10% of its activity when stored for a week in aliquots at -20°C. The same solutions stored in aliquots at 2 - 8°C. lose approximately 20% activity.

    1. Both of these products have the same specific activity (>2000 units/mg protein) for Dnase I. The enzyme impurity levels for each, however, are different. D5025 has the following impurity specification: Chymotrypsin (<0.5%), Protease (<0.05%), and RNase (<0.02%). The impurity specifications for D4527 are: Chymotrypsin (<0.01%), Protease (<0.005%), and RNase (<0.01%).

    1. DNA can be removed from preparations by incubation with 20 - 50 ug DNAse I in the presence of 50 mM Tris-HCl buffer, pH 7.5, 10 mM magnesium chloride, at 37°C. for 60 min.
