General description

  • Saggi enzimatici
  • Saggi basati sulle proteine
  • Tecniche sensibili alle proteasi che includono:
a. immunodosaggio enzimatico (EIA)
b. ibridazione di acidi nucleici
c. radioiimmunodosaggio (RIA)

Application

Features and Benefits

  • Frazionamento per shock termico
  • Priva di proteasi
  • Priva di acidi grassi
  • Essenzialmente priva di globuline

Preparation Note

Domande

1–10 di 20 domande
  1. What is the percentage composition of secondary structure?

    1 risposta
    1. The specific structural composition of this product is not determined on a lot-to-lot basis. However, this material is predominantly an α-helical protein. Based on available literature, the secondary structure consists of approximately 54–67% α-helix and 10–18% β-sheet or β-structure, with the remaining portion consisting of turns and unordered, or random-coil, regions. Variations may occur depending on the analytical method and experimental conditions.

      Utile?

  2. Is this BSA product (A7030) endotoxin free and suitable for cell culture usage?

    1 risposta
    1. This Bovine Serum Albumin is not tested for endotoxins. Please see the following options of BSA products which are tested for endotoxins:

      https://www.sigmaaldrich.com/product/sigma/a1933

      Utile?

  3. Would there be any concerns storing diluted aliquots at -70C for long term storage (1 year) or is it recommended still at 2-8C?

    1 risposta
    1. The stability of this material in solution, when stored at -70 °C, has not been evaluated. However, Albumin containing solutions are generally stable when stored at -20 to -80 °C in single use aliquots for at least 1 year. Also note that a number of sterile prepared BSA solutions currently available may be stored at 2–8 °C with assigned retest dates that range from 2 – 4 years from the date of Quality Release. Albumin is quite stable in solution and freezer storage may not be necessary.

      Utile?

  4. What is the concentration or percentage of fatty acid free?

    1 risposta
    1. The minimum allowable free fatty acid content is ≤ 0.02% (w/w), as determined by enzymatic assay. A review of recent lots indicates typical values of 0.00% free fatty acids. This information is lot-specific and reported in the Certificate of Analysis. Please see the link below to access a sample or lot-specific Certificate:

      Utile?

  5. Is this product biotin-free?

    1 risposta
    1. This product is not routinely tested for Biotin. However, this material is dialyzed with a 10 kDa cutoff membrane. This dialysis step removes small molecules such as biotin, which has a molecular weight of less than 500 Da. Therefore, this product is considered low in biotin, helping to prevent interference with streptavidin–biotin binding.

      Utile?

  6. If this material was inadvertently stored at room temperature, what is the room temperature stability?

    1 risposta
    1. This product is shipped at ambient temperature and is stable at ambient temperature for up to two weeks. For long term storage refrigerated temperature is recommended.

      Utile?

  7. How long would a 20% BSA solution be stable in water stored at 4C?

    1 risposta
    1. If stored at 2-8 °C, a filter sterilized 20% BSA solution in water or aqueous buffer is generally stable for a minimum of 2.5 - 3 years.

      Utile?

  8. Is it possible that BSA contains ions such as Mg2+? I am considering adding it to the buffer, but I am concerned that it might activate the enzyme.

    1 risposta
    1. The magnesium content of this material is not routinely tested, but historical data indicates that three lots contained between 10-20 parts per million. Water is used as the pre-lyophilization solvent.

      Utile?

  9. Is product number A7030, Bovine Serum Albumin (BSA) glycosylated?

    1 risposta
    1. Unfortunately, there is currently no information available regarding the glycosylation status of BSA product A7030.

      Utile?

  10. How does protease content determined? With this value all proteases are included? Or only refers to plasmatic proteases.

    1 risposta
    1. The specific method used to test this material for protease activity is considered proprietary. However, it is based on the enzymatic cleavage of FITC-Casein. See the link below to review the FITC-Casein Enzymatic Assay to demonstrate the principal of this method.
