Application
3,3′,5,5′-Tetramethylbenzidine has been used as a substrate for peroxidase in:
- enzyme-immunoassay (EIA)
- indirect competitive enzyme-linked immunosorbent assay (ELISA)
- sandwich enzyme-linked immunosorbent assay
La 3,3′,5,5′-tetrametilbenzidina (TMB) è un sostituto non cancerogeno (negativo al test di Ames) della benzidina adatto all′uso nelle procedure ELISA come substrato della perossidasi. Il substrato sviluppa un prodotto finale solubile di colore azzurro chiaro e può essere letto con spettrofotometria a 370 o 620-650 nm. La reazione della TMB può essere arrestata con 2 M di H2SO4 (dando luogo a una colorazione gialla) e può essere letta a 450 nm. Un reagente sensibile e specifico per il rilevamento della presenza di sangue, il saggio dell′emoglobina, il saggio della perossidasi.
