Specificity

Clone 1C18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects NEDD4-like E3 ubiquitin-protein ligase WWP2. It targets an epitope within the N-terminal half.

Immunogen

His-tagged recombinant fragment corresponding to 172 amino acids from the N-terminal half of human NEDD4-like E3 ubiquitin-protein ligase WWP2.

Application

Quality Control Testing

Evaluated by Western Blotting in C2C12 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected WWP2 in C2C12 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected WWP2 in C6 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected WWP2 in human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound recombinant WWP2 protein fragment with a KD of 3.0 x 10-7 in an affinity binding assay.

Target description

NEDD4-like E3 ubiquitin-protein ligase WWP2 (UniProt: O00308; also known as EC:2.3.2.26, Atrophin-1-interacting protein 2, AIP2, HECT-type E3 ubiquitin transferase WWP2, WW domain-containing protein 2) is encoded by the WWP2 gene (Gene ID: 11960) in human. WWP2 is a member of the C2-WW-HECT family (NEDD4 family) of the E3 ubiquitin ligases. It accepts ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then directly transfers the ubiquitin to targeted substrates. It is highly expressed in undifferentiated embryonic stem cells and its expression is reduced after embryoid body formation. It is detected in heart, throughout the brain, placenta, lung, liver, muscle, kidney, and pancreas. It is also detected in spleen and peripheral blood leukocytes. WWP2 has four WW domains that mediate its interaction with PPxY motif-containing proteins. It also has a C2 domain (aa 1-117) that is involved in autoinhibition of its catalytic activity by interacting with the HECT domain (aa 536-870). WWP2 is reported to regulate the pro-fibrotic gene network in primary cardiac fibroblasts and any defects in this gene can lead to pathologic cardiac fibrosis. It is also reported to regulate the nucleo-cytoplasmic shuttling and TGFβ-signal transduction activity of SMAD2. In untreated cardiac fibroblasts, it is present mainly in the cytoplasm, however, upon TGFβ1 stimulation it is localized predominantly in the nucleus. WWP2 is also serves as a physiological ubiquitin ligase for PTEN and promotes PTEN degradation under physiological conditions. WWP2 knockout mice exhibited reduced body size, elevated PTEN protein levels and antagonizes PI-3 kinase/Akt signaling. Four isoforms of WWP2 have been described that are produced by alternative splicing. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
