Human Caucasian prostate carcinoma

Cell Line Description

Derived from a metastasis at the left supraclavicular lymph node of a 50 year old patient with a confirmed diagnosis of metastatic prostate carcinoma. Growth and acid phosphatase production is affected by 5-alpha-dihydrotestosterone. They do not form a uniform monolayer and attach only lightly to the substrate. When shipped, cells detach from flask and can either be incubated 24-48 hours to allow attachment or be collected by centrifugation (150xg, 15 minutes) and reseeded.

Application

LNCap Clone FGC Cell Line has been used:
  • to study the effect of sex steroid hormones in regulating the expression of oligoadenylate synthetase 1 (OAS1)[1]
  • to study the role of curcumin and resveratrol in redox control and ability to promote apoptosis[2]
  • to investigate low molecular weight thiols present in the deproteinized extract of the cell line after derivatization[3]

Culture Medium

RPMI 1640 + 2mM Glutamine + 1.0 mM sodium pyruvate + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:6 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.i.e. seeding at 1-3 x 10,000 cells/cm2 Cells grow slowly in clusters and can be disaggregated by repeated pipetting. After subculture cells may take up to 48 hours to reattach. Do not disturb during this period. Medium is rapidly acidified, therefore change twice per week.

  1. Hi,I we are interested to purchase LNCap Clone FGC Cell Line human. My question is are they alos expressed PSMA?

    1 answer
    1. This cell line is a product of ECACC cell line. These cells are not tested for PSMA expression. However, there are a number of publications that suggest that prostate-specific membrane antigen is expressed. Please see the links below to review several examples:

    1. HPA Cultures does not perform any viral testing on the cell lines.

  5. How many Cell Line cells are in the vial?

    1 answer
    1. Each vial of cells contains 2-3 x 106 cells in 1 ml of freezing media. This is in a 1.8 mL ampule.  For exceptionally large cells, counts may decrease. Suspension cells, generally smaller cells, may contain as many as 4-5 x 106 cells/vial to assure optimal viability upon thaw.

  6. What medium should I use when I thaw the Cell Line cells?

    1. The medium for each cell line is listed on the product page. It is specific to each cell line. If not visible on the Sigma-Aldrich website product page, the HPA website also contains the same information (www.hpacultures.org.uk). The product number for the cell line is the same on either site.

  7. Why is Vapor Phase Liquid Nitrogen preferred for storage of Cell Line cells?

    1. If ampules are immersed into liquid phase of liquid nitrogen, it increases the risk of the liquid seeping into the vial. This could lead to problems of cross-contamination and increased risk of the ampule exploding when thawed.

  8. How do I handle Cell Line frozen cells upon arrival?

    1 answer
    1. Upon receipt, frozen ampules should be transferred directly to vapor phase liquid nitrogen without delay (-135°C) or liquid phase liquid nitrogen if vapor phase is not available. DO NOT use a -80°C freezer as an alternative; this will result in loss of viability.
