General description

CPNs are extremely bright and stable fluorophore nanoparticles which have been shown to give exceptional performance in a wide range of fluorescent analysis applications, such as lateral flow tests, fluorescent ELISA, cell tracking, immunofluorescence microscopy, flow cytometry and western blot. CPNs are resistant to photobleaching and a wide range of temperatures (-20°C to 120°C) and pH (2 to 12). They have been shown to be 1000x brighter than other fluorophores bringing exceptional levels of detection to assays and diagnostics.

Application

Amine Antibody Conjugation kit allows covalent linkage reactions to be readily carried out without access to specialist chemistry capabilities. The kit is designed to be specific for CPN – antibody / protein linkage has a rapid reaction time of 30mins and comes in three sizes for the conjugation of 10mcg, 30mcg and 100mcg of antibodies. CPNs are available in a range of emission wavelengths covering the visible and IR spectrum.

Questions

  1. Could you please let me know the CPN particle sizes, e.g., Cat#: 8400NG7 {fluorescence λem 840 nm (IR-I)}, and the particle-antibody conjugation ratios, with three sizes of antibodies (10mcg, 30mcg and 100mcg)? Thanks a lot!

    1 answer
    1. Product 8400NG5 is provided as one tube of 150 microliters each. Product 8400NG6 is provided as three tubes of 150 microliters each. Product 8400NG7 is provided as ten tubes of 150 microliters each. Each tube contains sufficient CPN (Conjugated Polymer Nanoparticles) to bind 10 micrograms of IgG antibody. So the amount of antibody (10 micrograms, 30 micrograms and 100 micrograms) corresponds to the amount of antibody that can be bound if using all of the tubes from the kit.

The CPN concentration is 0.1 mg/mL. Given 150 microliters per tube, that corresponds to 15 micrograms of CPN per tube. And if the contents of each tube can bind 10 micrograms of IgG antibody, that would give a CPN:antibody ratio of 3:2.

The particle size is 70-80 nm. It is described by the manufacturer as follows: "CPNs™ are water-soluble micelles compromising of a Light Emitting Polymer and are around 70-80 nm in size encapsulated within a biocompatible surfactant, increasing the hydrophilicity and allowing them to form micelles."
